Honor, Amongst Thieves is a Story Mission in Red Dead Redemption 2 (RDR2). This is one of the "Main Quests" of the game and players must complete it to advance the Story. Completing Story Missions opens up Stranger Missions as well as additional content throughout the game.
Honor, Amongst Thieves Mission Details
Colonel Favors is redirecting smallpox vaccine away from the Wapiti Reservation to punish the Native Americans for the recent unrest. Monroe requests Arthur's help to steal the vaccine from the army.

Recover The Vaccines
To start this mission, head back to Wapiti Indian Reserve and speak to Captain Monroe - keep in mind, you need to choose to help him as mentioned in Archeology for Beginners to unlock this mission.
After the cutscene, mount your horse and ride with the captain to the intercepted location. Once you've reached the vantage point, unmount your horse and follow Captain Monroe at the edge of the cliff to have a better view of where the wagon will be coming from - mount your horse, keep an eye on your map and follow the wagon.
You will have two ways to steal the vaccines - (1) you can choose to threaten the soldiers, start a gunfight and end up with killing them, or (2) you can sneak behind the wagon undetected, get on it and jump off once you've retrieved the vaccines. Whichever option you choose, you will need to get the item before the wagon reaches its destination.
It is all up to your preference, but it is recommended to recover the vaccines undetected and within the time limit to obtain a Gold Medal.
After stealing the vaccines, return back to Captain Monroe on horseback - another cutscene will trigger, which marks the completion of this mission.
