No, No and Thrice, No is a Story Mission in Red Dead Redemption 2 (RDR2). This is one of the "Main Quests" of the game and players must complete it to advance the Story. Completing Story Missions opens up Stranger Missions as well as additional content throughout the game.

Day Dreams Interrupted
Arthur Morgan (you) speaks to Mary-Beth Gaskill at the camp. She tells a bit about her past before asking you about your first encounter with Mary Linton. Before you can both get too much into your conversation, Susan Grimshaw storms towards you and tells you she requires your help in rescuing Tilly Jackson, as she's been kidnapped by the Foreman Brothers. You hop onto a wagon and ride out with Ms. Grimshaw.
Gritty Grimshaw
As you ride out, Grimshaw fills you in on the reason behind Tilly's disappearance also points out that Kieran Duffy has been missing for a while as well, and that she'd get a search team out to find him. When you arrive at the house, Ms. Grimshaw will talk to the guard momentarily before killing him with her knife. She then tells you to proceed into the house to find Tilly. Enter through the front door, and there will be a Foreman Brother -kill him. Then you'll find Tilly tied up and guarded by another Foreman, through the door on the left. Once you've killed him, free her and then give chase to the rest of the Foreman Brothers who ride off on their horses.
Ms. Grimshaw will mount the back of your horse as you go to take care of the final three Foreman Brothers who rode away. Kill the first two, then Lasso the third one. When you've done this, Ms. Grimshaw will take one of the dead men's horses back, allowing you to stow Anthony Foreman on the back of your horse. Bring him back to Tilly where after some talking, it is down to you to either kill him or spare him.
Whatever your decision may be, will complete the mission.
