Oh, Brother is a Stranger Mission in Red Dead Redemption 2 (RDR2). Stranger Missions are the equivalent of Side Quests, and they unlock as you progress the Story Missions.

Oh, Brother Walkthrough

Outside the general store in Valentine, there will be two brother fighting to prove who is tougher. Shoot the bottles on their heads after the cutscene.

Part II

Wait for a day, Proetus & Acrisius can be found in the backyard. This time, you are asked to punch them.

Part III

Wait for another day, they can be found in the west river of  New Hanover at Cumberland Falls. After pushing the barrels, ride with Helen to the highlighted area.
