A Fisher of Men is a Story Mission in Red Dead Redemption 2 (RDR2). This is one of the "Main Quests" of the game and players must complete it to advance the Story. Completing Story Missions opens up Stranger Missions as well as additional content throughout the game.

Favorite Uncle
Arthur Morgan (you) talks to Abigail Marston and she asks him if he can take Jack Marston fishing. You agree but as it's night time so you go bed and the next morning he talk to Jack about it. He's onboard too and grabs his fishing rod before jumping onto your horse. A bit of talk between the two before arriving by a stream. You unlock the Fishing Rod and are given a tutorial on how to fish.
Your first catch is too small to keep so you ponder throwing it back out. Whether you do, or not doesn't affect anything but a tiny bit of honor. Jack gets bored and instead sits down to play with some flowers.
Fisherman's Fisher
You get to practice your fishing a bit longer before you are interrupted by two men in bolo hats. They speak menacingly about knowing who you are and your affiliations with the Van Der Linde Gang before revealing themselves as Pinkerton agents, Jim Milton and Edgar Ross. They try to strike an offer with you to turn Dutch Van Der Linde in exchange for your freedom. Being the loyal buckerooni you are, you stand your ground and refuse the offer, so they leave.
Now that you've been rustled, you return to the camp with Jack. Once there, you tell Dutch about the confrontation and he says to lay low is the way forward.
